چکیده

Background It is thought that the interaction of genetic and environmental factors is an important risk factor for Parkinson’s disease (PD). a-Synuclein (a-syn) is a protein of special interest in PD because mutations in a-syn (A53T or A30P or E46K) lead to PD. Manganese (Mn) is a heavy metal known to cause parkinsonian symptoms. Therefore, we investigate the effect of manganese (Mn) on human a-syn-expressing mice.

Age-Dependent Effects of A53T Alpha-Synuclein on Behavior and Dopaminergic Function

Expression of A53T mutant human alpha-synuclein under the mouse prion promoter is among the most successful transgenic models of Parkinson's disease. Accumulation of A53T alpha-synuclein causes adult mice to develop severe motor impairment resulting in early death at 8-12 months of age. In younger, pre-symptomatic animals, altered motor activity and anxiety-like behaviors have also been reporte...

Dimebon Does Not Ameliorate Pathological Changes Caused by Expression of Truncated (1–120) Human Alpha-Synuclein in Dopaminergic Neurons of Transgenic Mice

BACKGROUND Recent clinical studies have demonstrated that dimebon, a drug originally designed and used as a non-selective antihistamine, ameliorates symptoms and delays progress of mild to moderate forms of Alzheimer's and Huntington's diseases. Although the mechanism of dimebon action on pathological processes in degenerating brain is elusive, results of studies carried out in cell cultures an...

Redistribution of DAT/α-Synuclein Complexes Visualized by “In Situ” Proximity Ligation Assay in Transgenic Mice Modelling Early Parkinson's Disease

Alpha-synuclein, the major component of Lewy bodies, is thought to play a central role in the onset of synaptic dysfunctions in Parkinson's disease (PD). In particular, α-synuclein may affect dopaminergic neuron function as it interacts with a key protein modulating dopamine (DA) content at the synapse: the DA transporter (DAT). Indeed, recent evidence from our "in vitro" studies showed that α-...

Disease-oriented Genome Networks Diseases of the Nervous System Network: Systematic Gene Identification and Functional Analyses in Common CNS Disorders Project: Functional Analyses in Genetic Mouse Model of Parkinson’s Disease

Introduction In 1997, based on linkage analyses of Italian and Greek families, alpha-synuclein was identified first as a gene involved in the pathogenesis of autosomal-dominantly inherited, early-onset Parkinson’s disease (PD). A nucleotide transition at position 209 from guanosine to adenosine (G209A), resulting in an alanine to threonine substitution at position 53 (A53T) on the amino acid le...

Absence of α-synuclein affects dopamine metabolism and synaptic markers in the striatum of aging mice☆

Despite numerous evidences for neurotoxicity of overexpressed alpha-synuclein, a protective function was suggested for endogenous alpha-synuclein and other members of the synuclein family. This protective role is most important for and evident in presynaptic terminals, where synucleins are normally accumulated. However, mice lacking synucleins display no adverse phenotype. In particular, no sig...
